PACAP and PAC1 Receptor Expression in Human Insulinomas

چکیده

Abstract Pituitary adenylate cyclase activating polypeptide (PACAP) is a neuropeptide with widespread occurrence and diverse functions. PACAP binds to specific PAC1 non-specific VPAC1/2 receptors. considered as growth factor, it plays important roles during development participates in reparative processes. Highest concentrations are found the nervous system endocrine glands, where several functions known, including actions tissue growth, differentiation tumour development. Therefore, we have investigated expression of its receptors different tumours, those glands. We showed earlier that receptor staining intensity decreased pancreatic ductal adenocarcinoma. In present study aimed investigate alterations human insulinoma compared immunostaining pattern samples from chronic pancreatitis patients. collected perioperative histological data patients who underwent operation because or over five-year-long period. Histology severe scar formation patients, while evidenced Grade 1 2 insulinoma. was studied using immunohistochemistry. Staining very strong Langerhans islets normal discernible also observed exocrine pancreas. Immunostaining for both markedly weaker samples, disappeared except intact islets. These findings show receptor/PACAP signalling altered this suggests possible involvement differentiation.

PACAP causes PAC1/VPAC2 receptor mediated hypertension and sympathoexcitation in normal and hypertensive rats.

Pituitary adenylate cyclase-activating polypeptide (PACAP) is an excitatory neuropeptide that plays an important role in hypertension and stress responses. PACAP acts at three G protein-coupled receptors [PACAP type 1 receptor (PAC(1)) and vasoactive intestinal peptide receptor types 1 and 2 (VPAC(1) and VPAC(2))] and is localized to sites involved in cardiovascular control, most significantly ...

Altered Circadian Food Anticipatory Activity Rhythms in PACAP Receptor 1 (PAC1) Deficient Mice

Light signals from intrinsically photosensitive retinal ganglion cells (ipRGCs) entrain the circadian clock and regulate negative masking. Two neurotransmitters, glutamate and Pituitary Adenylate Cyclase Activating Polypeptide (PACAP), found in the ipRGCs transmit light signals to the brain via glutamate receptors and the specific PACAP type 1 (PAC1) receptor. Light entrainment occurs during th...

Expression of the human PAC1 receptor leads to dose-dependent hydrocephalus-related abnormalities in mice.

Hydrocephalus is a common and potentially devastating birth defect affecting the CNS, and its relationship with G protein-coupled receptors (GPCRs) is unknown. We have expressed 2, 4, or 6 copies of a GPCR--the human PAC1 receptor with a 130-kb transgene in the mouse nervous system in a pattern closely resembling that of the endogenous gene. Consistent with PAC1 actions, PKA and PKC activity we...

PACAP-induced ERK activation in HEK cells expressing PAC1 receptors involves both receptor internalization and PKC signaling.

The pituitary adenylate cyclase-activating polypeptide (PACAP)-selective PAC1 receptor (Adcyap1r1) is a G protein-coupled receptor (GPCR) that activates adenylyl cyclase and PLC. Similar to many other GPCRs, our previous studies showed that the PAC1 receptor is internalized after ligand binding to form signaling endosomes, which recruit additional second messenger pathways. Using a human embryo...
